RESUMO

The ongoing Ebola virus outbreak in West Africa has highlighted questions regarding stability of the virus and detection of RNA from corpses. We used Ebola virus-infected macaques to model humans who died of Ebola virus disease. Viable virus was isolated <7 days posteuthanasia; viral RNA was detectable for 10 weeks.

RESUMO

Objective:To investigate the expression and postmortem stability of HSP70 in hippocampus of hemorrhagic shock rat.Methods:105 healthy SD rats were randomly divided into 5 groups:normal group,sham operation group,hemorrhagic shock group,traumatic control group and postmortem stability group.In hemorrhagic shock group and postmortem stability group,rats were submitted by controlled bleeding,with the mean arterial pressure kept around 40mmHg.And in traumatic group,brain contusion was induced by piston impact.After that,the regulation of HSP70 expression in hippocampus of each group was detected by immunohistochemistry technique,and evaluated with image analysis technique and statistic system.Results:One hour after hemorrhagic shock,HSP70 expression in hippocampus was significantly higher in each shock subgroups compared with that in normal group and sham group(P0.05).Conclusion:HSP70 is specifically expressed in hippocampus of hemorrhagic shock rat,and showed nice postmortem stability under the environment of -20℃.

Experimental acute myocardial ischemia model of dog was established, and the postmortem stability of fibronectin for the diagnosis of mycardial infarction was studied with immunohistochemistry and image analysis technique. The results showed that the positive reaction areas of fibronectin in ischemic myocardial tissues decreased along with the prolongation of postmortem interval, but positive reaction of fibronectin could still be found in ischemic myocardia kept for 4 weeks postmortem. No positive reaction for fibronectin could be found in normal myocardia when kept for different times. So fibronectin is a quite stable marker for the postmortem diagnosis of myocardial infarction, and is of practical value in the forensic practice.

Objective To observe the stability and the change pattern of VEGF expression in myocardium in varied postmortem periods,and to evaluate its usefulness in diagnosis of early myocardial ischemia after death.Methods An animal model of early myocardial ischemia was established by ligating coronary artery of rabbit,immunohistochemistry(SP protocol),image analysis technique and statistical analysis system are applied to detect and compare the area and intensity of expression of VEGF in cardiocytes,and its stability in different postmortem perieds.ResultsIn ischemic myocardium,VEGF shows foci or sheets of intense positive staining in cardiocytes,with negative or weak staining in some cells.There was weak staining in vascular endothelial cells and smooth muscle cells,but no staining in interstitium.When ischemic myocardium stored at 4℃,the staining of VEGF became weaker with time delay,but there was weak staining in a fraction of cardiocytes even 10d after death.There was significant difference in the intensity of VEGF staining between ischemic and normal postmortem myocardium from 1 to 10d.Conclusion VEGF can withstand a certain degree of autolysis or putrefaction,and the immunohistochemical staining of it is valuable in the diagnosis of early myocardial ischemia in corpses stored at 4℃ for 1~10d after death.
